Specimen Requirements
Preferred:
Container/Tube: Sterile Urine Container transferred into C&S preservative tube
Collection Instructions:
Whenever possible, collect specimen prior to administration of antibiotics because most
antibiotics concentrate in the urine. To avoid contamination of specimen with lower urethral flora,
voided specimens must be collected by a clean catch, midstream technique. Instructions are
given below:
Females
a. Wash hands thoroughly with soap and water.
b. With one hand, the patient is to spread her labia and keep them continuously
apart until the urine is voided.
c. Taking a towelette provided in the kit, wash vulva well, passing from front to
back.
d. Repeat the front to back washing 2 more times, using each of the towelettes
provided in the kit.
e. The patient should now void 20-25 ml of urine into the toilet to eliminate any
remaining skin flora. Collect the next portion of urine specimen in the container
provided with the kit (be careful not to touch the inside of the container).
f. Transfer 5.0 ml of urine into the urine vacutainer tube for culture and sensitivity. (see Resources tab for instructions.)
Males
a. Wash hands thoroughly with soap and water.
b. The foreskin must be completely retracted and the glans penis is cleansed with
the towelette provided.
c. The patient should void the first 20-30 ml of urine into the toilet. Collect the next
portion of urine into the container provided. Be careful not to touch the inside of
the container to avoid contamination.
d. Transfer 5.0 ml of urine into the urine vacutainer tube for culture and sensitivity. (see Resources tab for instructions)
